Sampling Methods

Samples were obtained using a sterile PVC corer, hammered into the ground. Approximately 50 g of soil were taken for each sample, and deposited in sterile, plastic zip-lock bags.

Study Extent Several soil samples were obtained on a single sampling event on Harderwijk, Gelderland, The Netherlands

Method step description:

  1. Soil samples were dried in a laminar flow cabinet. From each sample, 1 g of soil was serial diluted in sterile distilled water and different dilutions were plate in a variety of agar media. After incubation (aprox. 7 d, at room temperature), the plates were examined with the aid of a steromicroscope and pure cultures were made for each morphology observed. Pure cultures were individually stored and processed for identification by means of molecular data and phylogenetic analyses.
